| IL1RL1

Interleukin-1 Receptor-Like Protein 1, also known as IL1RL1 and ST2, is encoded by the IL1RL1 gene. Based on the function of its intracellular TIR domain, IL1RL1 is a member of the Toll-like receptor superfamily, but its extracellular region consists of immunoglobulin domains. Unlike other members of this family, IL1RL1 does not induce inflammatory responses by activating NF-κB, although it does activate MAP kinase. The gene was discovered on the long arm of chromosome 2 (2q12). It spans 40,536 bases and is located on the Watson (positive) strand. It encodes a 556-amino acid protein (molecular weight 63,358 Da). This protein is known to interact with MyD88, IRAK1, IRAK4, and TRAF6. It appears essential for the normal function of type 2 T helper cells (Th2 cells). ST2 is a member of the interleukin-1 receptor family. The ST2 protein exists in two isoforms directly implicated in the progression of cardiac disease: a soluble form (termed soluble ST2 or sST2) and a membrane-bound receptor form (termed ST2 receptor or ST2L).

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (pg/mL) 2000 1000 500 250 125 62.5 31.2 0
OD 2.22 1.56 0.92 0.63 0.42 0.25 0.18 0.08
Corrected OD 2.14 1.48 0.84 0.55 0.34 0.17 0.1 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Mouse IL1RL1 spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Serum(n=8) 84-101 96
Plasma(n=8) 92-105 102
Cell Culture Supernates(n=8) 96-108 105

| Linearity

Samples were spiked with high concentrations of Mouse IL1RL1 and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Serum(n=4) Plasma(n=4) Cell Culture Supernates(n=4)
1 : 2 Range (%) 84-95 88-96 90-110
Average (%) 91 93 96
